Standardisation explained: what 5% withanolides actually means

The percentage next to an extract is the most informative thing on a supplement label. What it measures, how it is verified, and what it does not promise.

Somewhere on a good supplement label you will see a phrase like "ashwagandha root extract, standardised to 5% withanolides". Most people read past it. It is the most informative thing on the pack.

What a standardisation percentage measures

A plant is not one substance. It is thousands, and only some of them are the compounds a formulator wants. Withanolides are the group of compounds in ashwagandha that research has focused on. Curcuminoids are the equivalent in turmeric. Tannins in amla.

An extract standardised to 5% withanolides has been processed and then tested so that withanolides make up 5% of its weight. In 300 mg of that extract, 15 mg is withanolides.

That is all the number means. It is a concentration, verified by testing, printed so you can check it.

Standardised extract vs plain herb powder

Compare two labels.

The first number is bigger. The second one is knowable. Raw root typically carries a far lower withanolide content than a standardised extract, and it varies by soil, season and supplier. Two bags of the same herb from the same farm can differ.

Standardisation is the step that removes that variation. It is why a 300 mg figure with a percentage next to it can be worth more than a 500 mg figure without one.

Why the plant part matters on an extract label

The plant part matters as much as the percentage. Ashwagandha research has largely used root extract. Leaf is cheaper and easier to source, and a label saying only "ashwagandha extract" is not telling you which one is in the tub.

Extract ratios like 10:1

The same applies elsewhere. Wheatgrass juice powder and wheatgrass leaf powder are different materials at different costs. A 10:1 moringa extract means roughly ten kilos of leaf went into one kilo of powder, which is a different product from dried leaf sold by the same name.

When a label names the plant but not the part or the ratio, the omission is usually deliberate.

How a standardisation figure is verified

The common method is HPLC, high performance liquid chromatography. A sample is separated into its components and the target compounds are measured against a known reference. A supplier issues a Certificate of Analysis for the batch showing the result.

You can ask any brand for the COA on the batch you were sent. Some will send it. What comes back tells you two things: whether they hold documentation per batch, and whether the number on the pack matches the number in the lab report.

A brand that publishes a percentage has committed to a figure that can be checked. That is the value of printing it, and the reason many brands do not.

What a standardisation percentage does not prove

This is where the reading has to stay honest.

A standardisation figure tells you what is in the pack. It does not tell you what the finished blend will do for you. Published research on an ingredient is research on that ingredient, usually at a specific dose, often in isolation, in a particular group of people. It is not a trial of a finished product that happens to contain it, and doses in a blend are frequently lower than doses in the studies people cite.

Anyone quoting ingredient research as though it were product research is skipping that step. The percentage on the label is a fact about the material. It is not a result.

Standardisation checks for any supplement label

  • Does each extract name the plant part, or just the plant?
  • Is a standardisation percentage printed, or only a weight?
  • Does the compound named match what research on that ingredient actually measures?
  • Is there a Certificate of Analysis available for your batch?
