Methylcobalamin, Methylfolate, D3: Active B Vitamins Explained

Active B vitamins are forms the body can use with fewer conversion steps: methylcobalamin, methylfolate, P-5-P. Why B12 and vitamin D shortfalls are common in India, what active forms change, and safe amounts.

Part of Ingredient evidence, graded. Related: Signal, a focus and clarity supplement.

Ajay Singh

Short answer: "active" B vitamins are forms that are already in, or close to, the shape the body uses: methylcobalamin for B12, L-methylfolate (5-MTHF) for folate, pyridoxal-5-phosphate (P-5-P) for B6 and riboflavin-5-phosphate for B2. They skip one or more conversion steps. For most people the standard forms work too; the active forms matter more for people who convert poorly. Separately, vitamin B12 and vitamin D shortfalls are common in India, especially among vegetarians and people who spend most of the day indoors.

Last updated: October 2026.

Key takeaways

  • Vegetarian diets supply little B12. Indian studies have found low B12 in a large share of adults.
  • Vitamin D deficiency is widespread in India despite the sun, because of indoor work, covered skin and pollution.
  • Active forms skip conversion steps. They are not "stronger"; they suit people who convert poorly.
  • Vitamin D3 (cholecalciferol) raises blood levels more effectively than D2 in most studies.
  • Upper limits exist. Do not stack several products without adding up the totals.

Active vs standard forms

Vitamin Standard form Active form
B12 Cyanocobalamin Methylcobalamin, adenosylcobalamin
Folate (B9) Folic acid L-methylfolate (5-MTHF)
B6 Pyridoxine hydrochloride Pyridoxal-5-phosphate (P-5-P)
B2 Riboflavin Riboflavin-5-phosphate
D D2 (ergocalciferol) D3 (cholecalciferol) is the form made in skin

Cyanocobalamin is stable, cheap and well studied, and the body converts it efficiently in most people. Methylcobalamin is the form most common in Indian prescriptions. Both correct B12 shortfall.

What about MTHFR?

MTHFR is an enzyme involved in converting folate to its active form. Common gene variants reduce its activity somewhat, and people with two copies of the main variant may convert folic acid less efficiently. L-methylfolate bypasses that step. Genetic testing is not needed to choose a supplement, and claims that MTHFR variants cause a long list of problems go well beyond the evidence.

Why are B12 and vitamin D low in India?

  • B12 comes almost only from animal foods. Dairy supplies some; a strict vegetarian or vegan diet supplies very little. Long-term metformin and acid-reducing medicine also lower B12 absorption. Why Indian diets need different nutrition.
  • Vitamin D is made in skin exposed to strong midday sun. Indoor work, covered clothing, darker skin, air pollution and few fortified foods mean many Indians test low.

If you think you are low, a blood test tells you. A diagnosed deficiency needs a measured dose from a doctor, often much higher than any daily blend provides.

How much do you need? Requirements and upper limits

Nutrient Adult requirement (ICMR-NIN 2020) Tolerable upper limit (NIH)
Vitamin B12 2.5 µg None set
Folate 300 µg (DFE) 1,000 µg from supplements
Vitamin B6 1.9 to 2.4 mg 100 mg
Vitamin D 600 IU (15 µg) 4,000 IU (100 µg)

Long-term high-dose B6 can cause nerve damage; problems have been reported below the formal limit in some people, so avoid stacking several B6-containing products. Vitamin D is fat-soluble and builds up, so add up every source you take.

Do B vitamins help focus and energy?

If you are deficient, correcting it can improve tiredness and, with B12, thinking. If you are not deficient, trials of B vitamins for energy or focus in healthy adults found small or no effects. They are a gap-filler, not a stimulant.

Who should check with a doctor first?

  • Anyone taking methotrexate, anti-seizure medicine or levodopa, which interact with folate or B6.
  • People with kidney disease, for vitamin D.
  • People with high blood calcium or sarcoidosis, for vitamin D.
  • Anyone pregnant or planning pregnancy: folate needs are higher, and the dose should come from your doctor.

What to check on the label

  • The form of each vitamin, and the amount in µg or mg, plus IU for vitamin D.
  • The percentage of your daily need, so you can spot megadoses.
  • Totals across everything you take.

Where SWATVA stands

Signal includes an active B-complex with vitamin D3, named in the ingredient ledger, at gap-filling amounts, not megadoses. Daily Greens has its own vitamin-mineral premix, so if you take both, check the combined totals in the ledger. Research on B vitamins does not establish an outcome for the finished blend, and Signal does not treat a deficiency.

B complex: what people search, answered

B complex benefits

B vitamins help turn food into energy, make red blood cells and keep nerves working. A B complex helps if you are short; it does not add energy above normal in people who are not.

When to take B complex: morning or night

Morning, with breakfast. Some people find late doses mildly stimulating.

B complex side effects

Bright yellow urine from B2 is harmless. Very high B6 over months can cause nerve symptoms, so avoid stacking products.

Common questions

Is methylcobalamin better than cyanocobalamin?

Both correct B12 shortfall. Cyanocobalamin is stable and converted efficiently by most people; methylcobalamin is already in an active form and is common in Indian prescriptions.

Why is vitamin D deficiency common in India?

Indoor work, covered clothing, darker skin, air pollution and few fortified foods mean many Indians make too little in their skin despite the sun.

Do B vitamins give you energy?

If you are deficient, correcting it can help tiredness. In healthy adults who are not deficient, trials found small or no effects on energy or focus.

When should I take a B complex?

In the morning with breakfast.

Does B complex give energy?

Only if you are short of B vitamins. In people who are not, trials found small or no effect.

Which SWATVA product has a B complex?

Signal carries an active B-complex with vitamin D3. Daily Greens carries B12 and vitamin D in its premix.
